If cells detect damage, the levels of these proteins go up and eventually stop the cells from dividing. This is not a bad thing, though; it is a cancer control mechanism. You don't want cells with damaged DNA to keep dividing! Of course knocking down p16 makes cells grow longer in culture. That doesn't mean that you want to knock down your own p16!
