[Effect of wuzi yanzong liquid on hypothalamus, monoamines, sexual
hormones and reproductivity in male rats]
About 100 male SD rats aged 2, 5, 10, 18 and 24 months (each age
group for about 20 rats) were used in the anti-senility experiment.
The animals of each age group were further subdivided into two groups
randomly: one group administered with Wuzi Yanzong Liquid (WYL) daily
for 5 weeks, and the other group with normal saline. After treatment,
plasma testosterone (T) and estradiol (E2), hypothalamus
norepinephrine (NE), dopamine (DA) and serotonin (5-HT) levels were
determined. In comparison with younger groups, the 18 and 24 months
old rats administered with saline had markedly lowered plasma T,
hypothalamus NE and DA levels, and elevated 5-HT, E2 levels and
5-HT/DA, E2/T ratios; while in the 18 and 24 months rats administered
with WYL the above mentioned changes were much less remarkable. Since
WYL only raised plasma T level in old rats but not in younger ones,
this drug seems to have different action from testosterone
preparations. In another animal experiment, the reproduction
promoting effect of WYL was shown by the increase of sperm count and
motility of sperms in old male rats as well as its reproductive ability.

[Clinical study on treatment of mild cognitive impairment by modified
wuzi yanzong granule]
OBJECTIVE: To observe the clinical efficacy and safety of modified
Wuzi Yanzong Granule (MWYG) in treating mild cognitive impairment
(MCI), and to explore its mechanism. METHODS: Forty-four MCI patients
were selected referring to the international recognized Peterson's
criteria and randomly divided into two groups, the treated group
treated with MWYG and the control group treated with Ginkgo leaf
extraction, with the course of 3 months for both groups. Changes of
memorial quotient (MQ), superoxide dismutase (SOD) activity,
malondialdehyde (MDA) content, mitochondrial DNA (mtDNA) deletion rate
and acetylcholinesterase (AchE) before and after treatment were
observed. RESULTS: After treatment, levels of MQ, serum SOD activity
increased and serum MDA content, mtDNA deletion rate and AchE decreased
in both groups (P < 0.01), but the difference between the two groups
was insignificant. No adverse reaction was found in two groups.
CONCLUSION: Both MWYG and Ginkgo leaf capsule can effectively improve
the memorial function of patients with MCI, the therapeutic mechanism
is possibly related with the actions in reducing AchE activity,
improving free radical metabolism, and alleviating mitochondrial DNA
oxidation damage.

[A clinical study of the effect of wuzi yanzong solution in
retarding aging process]
Eighty men ranging from 60 to 80 years old were selected, in whom
no disease was found through a thorough check-up except for some
senility manifestation diagnosed as deficiency of the kidney.
They were randomly divided into two groups, wuzi yanzong solution
(WYS) being administered in 50 cases and placebo in 30. After
5 weeks' treatment, symptoms such as hypomnsis, tinnitus, and
aching of the back and legs, dribbling after urinary voiding, and
nocturia were remarkably improved in the WYS group, but they
remained as before in the placebo group. Before the treatment,
plasma lipid peroxide (LPO), erythrocyte superoxide dismutase (SOD),
blood glutathione peroxidase (GSH-Px), serum luteinizing hormone
(LH) and follicle-stimulating hormone (FSH), plasma testosterone (T)
and estradiol (E2) levels were determined. In comparison with the
results obtained in 33 young men in their twenties, LPO, FSH, LH
level and E2/T ratio in the aged were greatly elevated, and SOD,
GSH-Px and T markedly lowered. After the treatment, there was
remarkable reduction of LPO and E2/T, and increase of SOD and T in
the WYS group, but no significant change in the placebo group. The
above results not only indicate the beneficial effect of WYS in
retarding the aging process, but also suggest the possible mechanism
of reducing the peroxidation of fatty acids by enhancing the
antioxidant enzymes to inhibit free radical activity.

Fufang wuzi yanzong pills have strong leukogenic effect on
cyclophosphamide induced leukopenia in mice. It can decrease the
lipid peroxide (MDA) content of liver in rats and lipofuscin
contents of heart and kidney in mice and increase the superoxide
dismutase (SOD) activity of liver in rats.

[Effect of wuzi yanzong Pill on mitochondrial DNA deletion and
respiratory chain enzyme complex activity in peripheral leukocyte of aged
male with kidney deficiency syndrome]
  OBJECTIVE: To investigate the effect of Wuzi Yanzong Pill (WZYZP) on
mitochondrial DNA (mtDNA) deletion and respiratory chain enzyme complex
(RCZC) in peripheral blood leukocyte of aged male with Kidney Deficiency
Syndrome (KDS). METHODS: Single-blinded study was conducted in 38 aged
male with KDS, who were randomly divided into 2 groups treated with WZYZP
and placebo respectively for 3 months. Levels of mtDNA deletion and RCZC
were determined by polymerase chain reaction (PCR) and enzyme kinetics
technique respectively. RESULTS: WZYZP could reduce the mtDNA deletion
and raise the activity of mitochondrial RCZC I, IV in peripheral blood
leukocyte of aged male with KDS (P < 0.05, P < 0.01). CONCLUSION: WZYZP
has protective effect on mtDNA from oxidative damage in leukocyte of aged
male with KDS.

[Effect of wuzi yanzong pill and its disassembled prescription on
mitochondrial DNA deletion, respiratory chain complexes and ATP synthesis
in aged rats]
  OBJECTIVE: To investigate the effect of Wuzi Yanzong Pill (WZYZP) and
its disassembled prescription on mitochondrial DNA deletions, respiratory
chain complexes and ATP synthesis in aged rats. METHODS: Animal
experiments with polymerase chain reaction (PCR), enzyme kinetics and
bioluminescence technique were conducted. RESULTS: WZYZP and its
disassembled prescription of Fructus Lycii and Semen Cuscutae could
reduce the mitochondrial DNA deletions and raise the activities of
mitochondrial respiratory chain complexes I, IV and adenosine
triphosphate (ATP) synthesis in aged rats' brain; Fructus Lycii and Semen
Cuscutae could also reduce the mitochondrial DNA deletions in aged rats'
heart (P < 0.05, P < 0.01). CONCLUSION: WZYZP, Fructus Lycii and Semen
Cuscutae have protective effect on oxidative damage of mitochondrial DNA
in aged rats.

Protective effect of Fructus Lycii polysaccharides against time and
hyperthermia-induced damage in cultured seminiferous epithelium.
  Lycium barbarum L. (Solanaceae) is a Chinese medicinal plant whose
fruits (Fructus Lycii) are used by Chinese physicians for treatment of
infertility. However, the active ingredients and the mechanism of action
underlying Lycium's fertility-facilitating effects remain unknown. Here
we report that Fructus Lycii polysaccharides (FLPS) inhibit time- and
hyperthermia-induced structural damage in murine seminiferous epithelium,
in vitro. Moreover, we found that FLPS delayed apoptosis in this system,
both at normothermic and hyperthermic culture conditions. Oxidative
stress was reported to be a major cause of structural degradation and
apoptosis in hyperthermic testes, and thus the protective effect of FLPS
could implicate an antioxidant mechanism of action. To test this
hypothesis we assayed the effect of FLPS on ultraviolet light-induced
lipid peroxidation, and cytochrome c reduction by free radicals. We found
that FLPS is a potent inhibitor of both of these reactions. Together,
these results demonstrate the protective effect of FLPS on time- and
hyperthermia-induced testicular degeneration in vitro, indicate the
potential mechanism of action for this protective effect, and provide
a scientific basis for the traditional use of this plant.

The inhibitory effect of extracts from Fructus lycii and Rhizoma
polygonati on in vitro DNA breakage by alternariol.
  Alternariol caused DNA single-strand breakage. Conversion of the closed
circular double-stranded supercoiled DNA (pBR 322) to the nicked circular
form and linear form was used to investigate the effect of extracts of
some Chinese medical herbs on DNA nicking induced by alternariol. Some
substances in the extracts of Rhizoma polygonati (RP) and Fructus lycii
(FL) were shown to protect DNA from the attack by alternariol. Some
substance in the RP may bind to plasmid DNA, and this binding reduces the
electrophoretic mobility of DNA. These results indicate that substances
from FL and RP may be used as DNA protectors. It is possible that they
play an important role in preventing cancer.
